O-156 Embryo-like models: Using 3D gastruloids to understand human development

چکیده

Abstract The mammalian embryo develops through coordinated cell fate decisions that enable emerging spatiotemporal and morphogenetic complexity. While much insight into this process has been gained studying model organisms, including mouse embryos, equivalent experiments are technically ethically challenging in the human embryonic context. Instead, we use gastruloids: 3-dimensional aggregates of pluripotent stem cells undergo multilineage differentiation to all three germ layers, polarise their gene expression exhibit axial organisation recapitulates many features gastrulation early organogenesis. By closely examining dynamics coordination spatial organisation, hope better understand regulatory logic behind developmental events, particularly those human-specific. Likewise, by carefully manipulating environment which these gastruloids grown, can bias structures towards particular lineages morphological interest. Doing so allows us explore principles development, provides an opportunity probe mechanisms biomedically-relevant conditions, such as congenital abnormalities.

Adjusting Animation Rigs to Human-Like 3D Models

In the animation process of a human-like 3D model, a skeleton must be specified to define the model’s surface deformation of its limbs. Nowadays the skeleton specification is hand made and very time consuming task. In this paper we propose a novel semi-automatic method for rigging a 3D model in an arbitrary pose using a skeleton defined in an animation datafile with no specific initial pose. Fi...

From cell death to embryo arrest: mathematical models of human preimplantation embryo development.

Human preimplantation embryos exhibit high levels of apoptotic cells and high rates of developmental arrest during the first week in vitro. The relation between the two is unclear and difficult to determine by conventional experimental approaches, partly because of limited numbers of embryos. We apply a mixture of experiment and mathematical modeling to show that observed levels of cell death c...

Using Multiple Models to Understand Data

A human’s ability to diagnose errors, gather data, and generate features in order to build better models is largely untapped. We hypothesize that analyzing results from multiple models can help people diagnose errors by understanding relationships among data, features, and algorithms. These relationships might otherwise be masked by the bias inherent to any individual model. We demonstrate this...

Using diffusion models to understand clinical disorders.

Sequential sampling models provide an alternative to traditional analyses of reaction times and accuracy in two-choice tasks. These models are reviewed with particular focus on the diffusion model (Ratcliff, 1978) and how its application can aide research on clinical disorders. The advantages of a diffusion model analysis over traditional comparisons are shown through simulations and a simple l...

Using Measurement Models to Understand Eyewitness Identification

Much research effort has been expended improving police lineup procedures used in collecting eyewitness identification evidence. Sequential presentation of lineup members, in contrast to simultaneous presentation, has been posited to increase witness accuracy, though analyses based in Signal Detection Theory (SDT) have challenged these claims. A possible way to clarify the effect of presentatio...
